Vitamin B1 has no catalytic activity on its own, so inside cells the enzyme TPK adds two phosphate groups, turning it into TPP, its working form.Vitamin B1 (thiamin) has no catalytic activity on its own. Absorbed from food (whole grains, lean pork, legumes, yeast) into blood and liver, it's modified inside cells by an enzyme called TPK (thiamin pyrophosphokinase) which adds two phosphate groups:· thiamin + → TPP (thiamin pyrophosphate) + AMP
TPP is the true working form of all B1 in the body — a common coenzyme / cofactor pattern.
What makes TPP special is the thiazole ring on the molecule — it carries an unusually acidic C atom (C2), easily deprotonated into a carbanion / ylide, which then nucleophilically attacks the α-C of ketone substrates.
This is the physical root of TPP's specifically cleaves α-keto acids chemistry — one small molecule, three big reaction families:
· PDH (pyruvate dehydrogenase) — glycolysis → entry
· α-KGDH (α-ketoglutarate dehydrogenase) — middle of TCA
· BCKDH (branched-chain α-keto acid dehydrogenase) — breakdown
· transketolase — pentose phosphate pathway
