Key facts:
· GCL is the rate-limiting enzyme for the entire GSH synthesis pathway.
· Glu is abundant in the cytoplasm (a crossroads of amino-acid metabolism), so it is not rate-limiting.
· Gly is also abundant, not rate-limiting.
· Cys is the rate-limiting substrate — dietary cysteine intake is low, the GSH system has large Cys demands, and Cys also participates in other pathways (taurine, methionine cycle).
· GCL is feedback-inhibited by GSH (excess GSH automatically shuts off further synthesis to prevent amino-acid waste).
Where enters: this is the exact physical position where NAC changes the story —
· After NAC enters the cell, an amidase removes the acetyl group → releases free Cys → raises the intracellular Cys pool.
· Cys pool rises → GCL's rate limit is lifted → γ-Glu-Cys synthesis rate rises → downstream GSH pool rises.
This is the single physical entry point for all of NAC's clinical utility (acetaminophen rescue / mucus / OCD adjunct).
