1 · Contract · Ca²⁺ release from SR
When an action potential reaches the T-tubule, the SR floods the cytoplasm with stored Ca²⁺, which binds troponin C and makes the fibre contract.Every step of muscle contraction is a story about calcium:· An action potential travels along the sarcolemma into the T-tubule
· L-type Ca²⁺ channels (DHPR) on the T-tubule sense the voltage and trigger the ryanodine receptors (RyR) on the sarcoplasmic reticulum (SR) membrane to open
· Ca²⁺ stored in the SR floods into the cytoplasm — cytosolic [Ca²⁺] rises 100-fold in milliseconds, from 100 nmol/L to 10 µmol/L
· Ca²⁺ binds troponin C (TnC) → unlocks the myosin-binding sites on actin → cross-bridge cycle → fibre slides and contracts.
